What dosages is glecaprevir/pibrentasvir available in?
glecaprevir/pibrentasvir is available as an oral tablets that contains 100 mg glecaprevir and 40 mg pibrentasvir and oral pellets that contains 50 mg glecaprevir and 20 mg pibrentasvir.
What is glecaprevir/pibrentasvir used for?
glecaprevir/pibrentasvir is an antiviral and is used for the following:
- Chronic (long-term) hepatitis C genotype 1, 2, 3, 4, 5, or 6 infection in certain people age 3 years and older
- Hepatitis C genotype 1 infection in people age 3 years and older who were previously treated with an NS5A inhibitor or an HCV protease inhibitor (not both)
How does glecaprevir/pibrentasvir work?
glecaprevir/pibrentasvir contains two antiviral medications that work together to stop the hepatitis C virus (HCV) from making copies of itself in the body. The two medications are glecaprevir (a protease inhibitor) and pibrentasvir (an NS5A inhibitor). They work by attacking two different proteins in the virus. Using two medications that work in different ways can prevent HCV resistance, which is when the virus develops a defense against antivirals. As a result, the medications no longer work to fight the virus.
How do I take glecaprevir/pibrentasvir?
glecaprevir/pibrentasvir is dosed as follows:
- Adults, children age 12 years or older, and children who weigh at least 99 pounds: The recommended dose is 3 tablets by mouth once a day with food. The total daily dose is 300 mg of glecaprevir and 120 mg of pibrentasvir
- Children age 3 to 12 years old: The dose is based on your child’s body weight. The provider will determine the right dose for your child. In general, the recommended dose ranges from 3 to 5 packets of oral pellets by mouth once a day with food
- The treatment duration ranges from 8 to 16 weeks, depending on your previous HCV treatment regimens and your liver health
Is it safe for me to take glecaprevir/pibrentasvir?
glecaprevir/pibrentasvir is a safe and effective treatment when used for FDA licensed indications. However, like all medications, they may give you unwanted side effects. You should always discuss potential side effects with your physician to ensure the medication is suitable and right for you. You should not use this medication in the following:
- Patients with moderate or severe hepatic impairment (Child-Pugh B or C) or those with any history of prior hepatic decompensation
- Coadministration with atazanavir or rifampin

Common side effects of glecaprevir/pibrentasvir:
- Headache
- Fatigue or tiredness
- Nausea
glecaprevir/pibrentasvir Serious Side Effects
Serious side effects are rare with glecaprevir/pibrentasvir. Contact your healthcare provider immediately if you experience any of the following.
- Liver failure caused by hepatitis B virus reactivation: nausea, stomach pain, yellowing of your skin or whites of your eyes
Effects of other drugs, drug classes and over-the-counter products on glecaprevir/pibrentasvir
The following medications may interact with glecaprevir/pibrentasvir:
- Carbamazepine, efavirenz, and St. John’s wort may decrease concentrations of glecaprevir and pibrentasvir. Coadministration of carbamazepine efavirenz containing regimens, and St. John’s wort with glecaprevir/pibrentasvir is not
- recommended
- Clearance of HCV infection with direct-acting antivirals may lead to changes in hepatic function, which may impact safe and effective use of concomitant medications. Frequent monitoring of relevant laboratory parameters (INR or blood glucose) and dose adjustments of certain concomitant medications may be necessary
- Medication- Assisted Treatment (MAT) for Opioid Use Disorder
- Consult the full prescribing information prior to and during treatment for potential drug interactions
