Prempro is an estrogen replacement / progestin and is used for the following:
- Hot flashes, night sweats, and flushes caused by menopause
- Vaginal dryness and burning caused by menopause
- Prevention of postmenopausal osteoporosis (not a first-choice)
How does Prempro work?
Prempro is a combination of two medications. Conjugated estrogens is a mixture of estrogens that replaces the estrogen your body loses during menopause. It relieves hot flashes, relieves vaginal dryness, and strengthens bones. Medroxyprogesterone is a progestin that protects the uterus from damage and helps prevent uterine cancer.
How do I take Prempro?
Prempro: Take 1 tablet by mouth once daily. Since there are many strengths of conjugated estrogens and medroxyprogesterone available, ask your provider which combination strength you should be taking.
Is it safe for me to take Prempro?
Prempro is a safe and effective treatment when used for FDA licensed indications. However, like all medications, they may give you unwanted side effects. You should always discuss potential side effects with your physician to ensure the medication is suitable and right for you. You should not use this medication if any of the following apply:
- Undiagnosed abnormal genital bleeding
- Known, suspected, or history of breast cancer
- Known or suspected estrogen-dependent neoplasia
- Active DVT, PE, or a history of these conditions
- Active arterial thromboembolic disease (for example, stroke and MI), or a history of these conditions
- Known anaphylactic reaction oar ngioedema to Prempro/PREMPHASE
- Known liver dysfunction or disease
- Known protein C, protein S, or antithrombin deficiency, or other known thrombophilic disorders.
- Known or suspected pregnancy

Common side effects of Prempro:

- More vaginal discharge than usual
Prempro Serious Side Effects
Serious side effects are rare with Prempro. Contact your healthcare provider immediately if you experience any of the following.
- Allergic reactions
- Heart attack: chest pain or tightness, pain that radiates up to your shoulder, arm, neck, or jaw
- Stroke: sudden trouble talking, trouble walking, or numbness on one side of your body or face
- Shortness of breath
- Sudden, severe headaches
- Breast lumps or pain
- Unusual or heavy vaginal bleeding
Effects of other drugs, drug classes and over-the-counter products on Prempro
The following medications may interact with Prempro:
- Inducers and/or inhibitors of CYP3A4 may affect estrogen drug metabolism
- Aminoglutethimide administered concomitantly with MPA may significantly depress the bioavailability of medroxyprogesterone acetate
