PREMPHASE is the brand name for conjugated estrogens/medroxyprogesterone acetate and is available as an oral tablet.

PREMPHASE is available as an oral tablet that contains 0.625 mg CE and 0.625 mg CE plus 5 mg MPA.
What is PREMPHASE used for?
PREMPHASE is an estrogen replacement / progestin and is used for the following:
- Hot flashes, night sweats, and flushes caused by menopause
- Vaginal dryness and burning caused by menopause
- Prevention of postmenopausal osteoporosis (not a first-choice)
How does PREMPHASE work?
PREMPHASE is a combination of two medications. Conjugated estrogens is a mixture of estrogens that replaces the estrogen your body loses during menopause. It relieves hot flashes, relieves vaginal dryness, and strengthens bones. Medroxyprogesterone is a progestin that protects the uterus from damage and helps prevent uterine cancer.
How do I take PREMPHASE?
PREMPHASE: There are 2 different colored tablets in the package. Take 1 dark red tablet by mouth once daily on days 1 through 14. Take 1 light blue tablet by mouth once daily on days 15 through 28. The dark-red tablet contains 0.625 mg of conjugated estrogens only, and the light blue tablet contains 0.625 mg conjugated estrogens and 5 mg of medroxyprogesterone.
Is it safe for me to take PREMPHASE?
PREMPHASE is a safe and effective treatment when used for FDA licensed indications. However, like all medications, they may give you unwanted side effects. You should always discuss potential side effects with your physician to ensure the medication is suitable and right for you. You should not use this medication if any of the following apply:
- Undiagnosed abnormal genital bleeding
- Known, suspected, or history of breast cancer
- Known or suspected estrogen-dependent neoplasia
- Active DVT, PE, or a history of these conditions
- Active arterial thromboembolic disease (for example, stroke and MI), or a history of these conditions
- Known anaphylactic reaction or angioedema to PREMPRO/PREMPHASE
- Known liver dysfunction or disease
- Known protein C, protein S, or antithrombin deficiency, or other known thrombophilic disorders
- Known or suspected pregnancy
PREMPHASE Common Side Effects
Common side effects of PREMPHASE:
- Breast pain
- Headaches
- Stomach pain
- Menstrual cramps
- Gas
- Nausea
- Depression
- More vaginal discharge than usual
PREMPHASE Serious Side Effects
Serious side effects are rare with PREMPHASE. Contact your healthcare provider immediately if you experience any of the following.
- Allergic reactions
- Heart attack: chest pain or tightness, pain that radiates up to your shoulder, arm, neck, or jaw
- Stroke: sudden trouble talking, trouble walking, or numbness on one side of your body or face
- Shortness of breath
- Sudden, severe headaches
- Breast lumps or pain
- Unusual or heavy vaginal bleeding
Effects of other drugs, drug classes and over-the-counter products on PREMPHASE
The following medications may interact with PREMPHASE:
- Inducers and/or inhibitors of CYP3A4 may affect estrogen drug metabolism
- Aminoglutethimide administered concomitantly with MPA may significantly depress the bioavailability of medroxyprogesterone acetate
