How does BRAFTOVI work?
BRAFTOVI is a kinase inhibitor known as a BRAF inhibitor. The BRAF protein is part of a whole chain of proteins that help cells (including cancer cells) grow. When the BRAF protein is mutated, the cancer grows uncontrollably and can spread more quickly. By blocking BRAF, BRAFTOVI helps stop tumor growth.
How do I take BRAFTOVI?
Melanoma: The typical dose is 450 mg (six 75 mg capsules) by mouth once daily. Colorectal cancer: The typical dose is 300 mg (four 75 mg capsules) by mouth once daily.

BRAFTOVI is a safe and effective treatment when used for FDA licensed indications. However, like all medications, they may give you unwanted side effects. You should always discuss potential side effects with your physician to ensure the medication is suitable and right for you.

Common side effects of BRAFTOVI:
- Fatigue
- Nausea
- Lower red blood cells
- Vomiting
- Stomach pain
- Higher liver enzymes in blood

- Lower white blood cell count

- Constipation
- Rash
- Headache
BRAFTOVI Serious Side Effects
Serious side effects are rare with BRAFTOVI. Contact your healthcare provider immediately if you experience any of the following.
- Bleeding: blood in stool, bleeding from rectum, sudden headache
- Changes in heart rhythm: lightheadedness, fainting, seizures, loss of consciousness
Effects of other drugs, drug classes and over-the-counter products on BRAFTOVI
The following medications may interact with BRAFTOVI:
- Strong or moderate CYP3A4 inhibitors: Avoid coadministration. If unavoidable, reduce BRAFTOVI dosage
- Strong CYP3A4 inducers: Avoid coadministration.
- Sensitive CYP3A4 substrates: Avoid coadministration with CYP3A4 substrates (including hormonal contraceptives) for which a decrease in plasma concentration may lead to reduced efficacy of the substrate
- Transporters: Dose reductions of drugs that are substrates of OATP1B1, OATP1B3, or BCRP may be required when used concomitantly with BRAFTOVI

Is BRAFTOVI safe in pregnancy?
BRAFTOVI can harm your unborn baby. Females who are able to become pregnant should use effective non-hormonal birth control (contraception) during treatment with BRAFTOVI and for 2 weeks after the last dose of BRAFTOVI. Birth control methods that contain hormones (such as birth control pills, injections or transdermal systems) may not work as well during treatment with BRAFTOVI. Talk to your healthcare provider about birth control methods that may be right for you during this time. Your healthcare provider will do a pregnancy test before you start taking BRAFTOVI. Tell your healthcare provider right away if you become pregnant or think you might be pregnant during treatment with BRAFTOVI.
