Encorafenib is a BRAF inhibitor and is used for the following:
Melanoma with certain BRAF mutations that has spread (metastatic) or that can’t be removed by surgery, used with Mektovi (binimetinib) Colorectal cancer with a certain BRAF mutation that has spread (metastatic), used with Erbitux (cetuximab)
How does Encorafenib work?
Encorafenib is a kinase inhibitor known as a BRAF inhibitor. The BRAF protein is part of a whole chain of proteins that help cells (including cancer cells) grow. When the BRAF protein is mutated, the cancer grows uncontrollably and can spread more quickly. By blocking BRAF, Encorafenib helps stop tumor growth.
How do I take Encorafenib?
Melanoma: The typical dose is 450 mg (six 75 mg capsules) by mouth once daily. Colorectal cancer: The typical dose is 300 mg (four 75 mg capsules) by mouth once daily.

- Changes in heart rhythm: lightheadedness, fainting, seizures, loss of consciousness
Effects of other drugs, drug classes and over-the-counter products on Encorafenib
The following medications may interact with Encorafenib:
- Strong or moderate CYP3A4 inhibitors: Avoid coadministration. If unavoidable, reduce Encorafenib dosage
- Strong CYP3A4 inducers: Avoid coadministration.
- Sensitive CYP3A4 substrates: Avoid coadministration with CYP3A4 substrates (including hormonal contraceptives) for which a decrease in plasma concentration may lead to reduced efficacy of the substrate
- Transporters: Dose reductions of drugs that are substrates of OATP1B1, OATP1B3, or BCRP may be required when used concomitantly with Encorafenib

Is Encorafenib safe in pregnancy?
Encorafenib can harm your unborn baby. Females who are able to become pregnant should use effective non-hormonal birth control (contraception) during treatment with Encorafenib and for 2 weeks after the last dose of Encorafenib. Birth control methods that contain hormones (such as birth control pills, injections or transdermal systems) may not work as well during treatment with Encorafenib. Talk to your healthcare provider about birth control methods that may be right for you during this time. Your healthcare provider will do a pregnancy test before you start taking Encorafenib. Tell your healthcare provider right away if you become pregnant or think you might be pregnant during treatment with Encorafenib.
