What is Vonjo used for?
Vonjo is a type of targeted therapy called a JAK inhibitor. It’s used to treat adults with a rare blood cancer called myelofibrosis (MF).
How does Vonjo work?
Vonjo is is a JAK inhibitor, which means it blocks JAK2 proteins. These proteins usually send signals that tell blood cells to grow and divide. The medication can also block the protein IRAK1, which does something similar. People with MF have abnormal JAK2 proteins that work out of control. By blocking these abnormal JAK2 proteins, Vonjo helps stop abnormal blood cells from growing too much in people with MF.
How do I take Vonjo?
The typical dose of Vonjo is 200 mg by mouth twice a day.
Is it safe for me to take Vonjo?
Vonjo is a safe and effective treatment when used for FDA licensed indications. However, like all medications, they may give you unwanted side effects. You should always discuss potential side effects with your physician to ensure the medication is suitable and right for you. You should not use this medication in the following:
- Concomitant use of strong CYP3A4 inhibitors or inducers

Common side effects of Vonjo:
- Diarrhea
- Low platelets
- Nausea
- Anemia
- Swelling in the hands, ankles, or feet
- Vomiting
- Dizziness
- Fever
Vonjo Serious Side Effects
Serious side effects are rare with Vonjo. Contact your healthcare provider immediately if you experience any of the following:
- Bleeding: bruising that you can’t explain, bleeding that doesn’t stop, getting nosebleeds often, blood in urine, blood in stool
- Changes in heart rhythm: lightheadedness, blurry vision, fainting, passing out
- Blood clot: swelling in one arm or one leg, sudden and severe chest pain
- Heart attack: chest pain that spreads to the arm or jaw, pressure in the chest, sweating, shortness of breath, nausea
- Stroke: sudden and severe headache, weakness on one side of the body, drooping face, trouble speaking, trouble seeing, trouble walking, confusion
- Infection: a body temperature of 100.4ºF or higher, chills, body aches, cough

The following medications may interact with Vonjo:
- Substrates of P-gp, BCRP, and CYP1A2
- Inducers of inhbitors of CYP3A4 and CYP2C19
