What is SCEMBLIX used for?
SCEMBLIX is a tyrosine kinase inhibitor and is used to treat Philadelphia-positive chronic myelogenous leukemia (Ph+ CML).
How does SCEMBLIX work?
SCEMBLIX is a tyrosine kinase inhibitor (TKI). It blocks a certain protein in your body called BCR-ABL tyrosine kinase. This protein is found on cancer cells like CML and not on normal, healthy cells. It causes the cancer cells to grow and multiply. By blocking this protein, SCEMBLIX kills cancer cells and stops their spread.
How do I take SCEMBLIX?
The typical starting dose of SCEMBLIX is 80 mg taken by mouth once daily. Another recommended option is to take 40 mg twice daily, about 12 hours apart. If you have the T315I mutation in your cancer, the recommended starting dose is 200 mg taken by mouth twice daily, about 12 hours apart.

SCEMBLIX is a safe and effective treatment when used for FDA licensed indications. However, like all medications, they may give you unwanted side effects. You should always discuss potential side effects with your physician to ensure the medication is suitable and right for you.
SCEMBLIX Common Side Effects
Common side effects of SCEMBLIX:
- Bone or muscle pain

SCEMBLIX Serious Side Effects
Serious side effects are rare with SCEMBLIX. Contact your healthcare provider immediately if you experience any of the following.
- Heart or blood vessel problems: shortness of breath; chest pain; fast heartbeat; swelling in your ankles or feet; numbness on one side of your body; vision changes; severe stomach area pain; pain in your arms, legs, back, neck, or jaw
Effects of other drugs, drug classes and over-the-counter products on SCEMBLIX
The following medications may interact with SCEMBLIX:
- Strong CYP3A4 Inhibitors: Closely monitor for adverse reactions during concomitant use of SCEMBLIX at 200 mg twice daily.
- Itraconazole Oral Solution Containing Hydroxypropyl-β-cyclodextrin: Avoid concomitant use of SCEMBLIX at all recommended doses
- Certain Substrates of CYP3A4: Closely monitor for adverse reactions during concomitant use of SCEMBLIX at 80 mg total daily dose. Avoid use of SCEMBLIX at 200 mg twice daily
- Substrates of CYP2C9: Avoid concomitant use of SCEMBLIX at all recommended doses
- 80 mg total daily dose: If unavoidable, reduce the CYP2C9 substrate dosage as necessary
- 200 mg twice daily: If unavoidable, consider alternative therapy with non-CYP2C9 substrate
- Certain P-gp Substrates: Closely monitor for adverse reactions during concomitant use of SCEMBLIX at all recommended doses
- Substrates of OATP1B or BCRP: Avoid concomitant use with rosuvastatin and atorvastatin at all recommended doses. Closely monitor for adverse reactions of other OATP1B or BCRP substrates during concomitant use of

Is SCEMBLIX safe in pregnancy?
SCEMBLIX can harm your unborn baby. Your healthcare provider will do a pregnancy test before you start treatment with SCEMBLIX. Females who are able to become pregnant should use effective birth control during treatment and for 1 week after your last dose of SCEMBLIX. Talk to your healthcare provider about birth control methods that may be right for you. Tell your healthcare provider right away if you become pregnant or think you may be pregnant during treatment with SCEMBLIX.
