Erdafitinib is prescribed for the treatment of urothelial carcinoma, a type of bladder cancer that has either metastasized or is inoperable.

Adverse reactions reported in 10% or more of patients (any grade) or 5% or more of patients (grade 3-4) included eye disorders such as dry eye in 286 patients, blurred vision in 170 patients, and increased lacrimation in 100 patients.

As of January 15, 2023, patients who were treated with Balversa had an overall survival rate of 12.1 months, whereas patients who received chemotherapy had an overall survival rate of 7.8 months.

During treatment for WM, patients commonly experienced fatigue (72%), neutropenia (47%), nausea (39%), anemia (37%), peripheral neuropathy (37%), thrombocytopenia (30%), rash and itch (26%), back and joint pain (23%), mouth sores (22%), diarrhea (20%), headache (19%), and hair loss (17%).

The mechanism of action of Balversa (erdafitinib) involves inhibiting the enzymatic activity of fibroblast growth factor receptors (FGFR1, FGFR2, FGFR3, and FGFR4) by binding to them. Additionally, erdafitinib also binds to other receptors such as RET, CSF1R, PDGFRA, PDGFRB, FLT4, KIT, and VEGFR2.

Patients were given pemigatinib in 21-day cycles, taking 13.5 mg orally once daily for 14 days, followed by 7 days off treatment. This continued until disease progression or unacceptable toxicity.

The initial dose of BALVERSA® is 8 mg (equivalent to two 4 mg tablets) taken orally once a day. After 14 to 21 days, the dose may be increased to 9 mg (equivalent to three 3 mg tablets) once a day depending on serum phosphate levels and how well the patient tolerates the medication. The research plan anticipated a 46% enhancement in the median overall survival (OS) of the erdafitinib group compared to the pembrolizumab group, with a hazard ratio (HR) of 0.69 for erdafitinib in relation to the pembrolizumab group, assuming an exponential distribution.

This approval allows for the use of motixafortide in combination with filgrastim to mobilize hematopoietic stem cells to the peripheral blood. FGFR mutation or overamplification has been demonstrated to be responsible for the development of breast, lung, endometrial, gastric cancers, and hepatocellular carcinoma. At present, there are three FGFR inhibitors that have received FDA approval: pemigatinib, futibatinib, and infigratinib.
