Apretude is an antiretroviral that is used as HIV pre-exposure prophylaxis (PrEP).
How does Apretude work?
Apretude is an integrase inhibitor. It works by blocking an enzyme (protein) called integrase that HIV uses to insert its genetic material into your cells. By stopping this step, the virus can’t cause an infection in your body.
How do I take Apretude?
Apretude is typically dosed as follows:
- The usual starting dose is one 600 mg injection into the buttock muscle once a month for 2 months.
- The maintenance dose is one 600 mg injection once every 2 months thereafter.
Is it safe for me to take Apretude?
Apretude is a safe and effective treatment when used for FDA licensed indications. However, like all medications, they may give you unwanted side effects. You should always discuss potential side effects with your physician to ensure the medication is suitable and right for you. You should not use this medication in the following:
- Are HIV positive or have an unknown HIV status
- Take certain seizure medications like carbamazepine, oxcarbazepine, phenobarbital, and phenytoin
- Take certain antibiotics like rifampin and rifapentine

Common side effects of Apretude:
- Injection site reaction
- Headache
- Diarrhea
- Nausea
- Dizziness
- Fever
- Upper respiratory tract infection
Apretude Serious Side Effects
Serious side effects are rare with Apretude. Contact your healthcare provider immediately if you experience any of the following:
- Serious allergic reactions: rash, mouth sores, feeling ill, trouble breathing, tiredness, fever, muscle or joint pain, swelling in the face, redness or swelling in the eyes
- Liver problems: upper right stomach pain, yellowing of the skin or eyes, dark urine, light-colored stools nausea, vomiting, reduced appetite, itching
- Mood changes: feeling sad, restlessness, anxiety, suicidal thoughts and behavior

The following medications may interact with Apretude:
- Carbamazepine
- Oxcarbazepine
- Phenobarbital
- Phenytoin
- Rifampin

It is not known whether this medication is safe for you and your unborn baby during pregnancy. Talk to your doctor if you are already pregnant or if you have plans on becoming pregnant. They will recommend what is best for you. There is a pregnancy exposure registry that monitors pregnancy outcomes in individuals exposed to Apretude during pregnancy. Healthcare providers are encouraged to register their patients if they take this medication while pregnant.
