Elagolix is a gonadotropin-releasing hormone (GnRH) antagonist, meaning that it attaches to and blocks GnRH receptors (a protein that responds to signals) in your pituitary gland. This lowers the release of hormones that cause pain due to endometriosis.
How do I take Elagolix?
Elagolix is typically started at 150 mg by mouth once daily for 24 months. Your dose might differ if you have dyspareunia (genital pain during sex) or liver problems.
Is it safe for me to take Elagolix?
Elagolix is a safe and effective treatment when used for FDA licensed indications. However, like all medications, they may give you unwanted side effects. You should always discuss potential side effects with your physician to ensure the medication is suitable and right for you. You should not use this medication if any of the following apply.
- Pregnancy
- Known osteoporosis
- Severe hepatic impairment
- Organic anion transporting polypeptide (OATP) 1B1 inhibitors that significantly increase Elagolix plasma concentrations

- Serious allergic reaction: rash, swelling of lips, tongue, or throat, difficulty breathing
Effects of other drugs, drug classes and over-the-counter products on Elagolix
The following medications may interact with Elagolix:
- Increase monitoring of digoxin concentrations and potential signs and symptoms of clinical toxicity when initiating Elagolix in patients who are taking digoxin If Elagolix is discontinued, increase monitoring of digoxin concentrations.
- Monitor lipid levels and adjust the dose of rosuvastatin, if necessary
- Advise women to use effective non-hormonal contraception during treatment with Elagolix and for 28 days after discontinuing Elagolix
- Concomitant use of Elagolix 200 mg twice daily and strong CYP3A inhibitors for more than 1 month is not recommended. Limit concomitant use of Elagolix 150 mg once daily and strong CYP3A inhibitors to 6 months
- Co-administration of Elagolix with strong CYP3A inducers may decrease Elagolix plasma concentrations and may result in a decrease of the therapeutic effects of Elagolix
- Concomitant use of Elagolix 200 mg twice daily and rifampin is not recommended. Limit concomitant use of Elagolix 150 mg once daily and rifampin to 6 months
